Tissue Fracture Toughness of Bone Measrued by Microindentation

Introduction: The fracture mechanisms of bone tissue have been described as a damage accumulation process, which starts with the initiation and propagation of microcracks. Knowledge of crack initiation and propagation at tissue level (tissue fracture toughness) is critical to understand and prevent bone fractures. Tissue fracture toughness could provide valuable information for defining bone quality, evaluating efficacy of drug treatment, and revealing tissue deterioration due to aging or disease. In this study, we designed a novel microindentation testing method to force pre-existing naturally occurring linear microcracks to open. Based on the recorded load and displacement data and the measurement of crack opening length, the crack initiation and propagation process can be identified and monitored. An indentation fracture mechanics model was used to calculate the fracture toughness. The results showed that our microindentation testing protocol can be used to study the bone tissue fracture mechanism and provide fracture toughness measurement.
